### 1. Overview of Peptide Lyophilization Lyophilization (freeze-drying) removes water from synthesized peptide solutions under high vacuum and sub-zero temperatures. This process places the polypeptide chain in an amorphous crystalline state that drastically suppresses hydrolytic and enzymatic degradation pathways. ### 2. Selection of Reconstitution Solvents - **Bacteriostatic Water (0.9% Benzyl Alcohol)**: Optimal for multi-aliquot laboratory protocols extending over multiple days/weeks. The benzyl alcohol inhibits microbial contamination. - **Sterile 0.9% NaCl (Saline)**: Recommended for sensitive bioassays where benzyl alcohol might interfere with cellular assays. - **Dilute Acetic Acid (0.1%-1%) or Ammonium Hydroxide**: Necessary for hydrophobic peptides with extreme isoelectric points (e.g. basic or acidic residues). ### 3. Reconstitution Step-by-Step Procedure 1. Allow the lyophilized vial and diluent to equilibrate to ambient room temperature (20-25°C) for 15-20 minutes to prevent condensation shock. 2. Clean rubber septum stoppers with sterile 70% isopropanol swabs. 3. Using a sterile laboratory syringe, slowly draw the calculated volume of diluent (e.g. 1.0 mL to 3.0 mL). 4. Direct the needle stream against the inner glass wall of the vial—**do NOT squirt directly onto the powder cake**. 5. Gently swirl the vial in circular motions until the lyophilized cake is fully dissolved. **Never shake vigorously**, as excessive shearing force can cause denaturing or aggregation of tertiary peptide chains. ### 4. Cold-Chain Storage Guidelines - **Lyophilized Powder**: Store at -20°C for up to 36 months. Protect from direct ultraviolet light. - **Reconstituted Solution**: Aliquot immediately into sterile microcentrifuge tubes to prevent repeated freeze-thaw cycles. Store refrigerated at 2°C–8°C for 21–30 days or frozen at -80°C for prolonged research storage.
